BIOMIMESYS® plates should be taken out from the fridge (4°C) and unpacked just before use.
The cells should be seeded within a small volume of medium (20-30 µL per hydroscaffold), carefully loaded at the surface in the middle of each hydroscaffold, without touching the matrix with the pipette tip. The drop containing cells will immediately be absorbed, and the cells will enter the pores of the matrix: from white (dehydrated hydroscaffold), BIOMIMESYS® will get translucent. When you have seeded all hydroscaffolds in the 96-well plate, fill each well with 200 µL of medium and incubate the cells in your incubator as usual (ex: 37°C, 5% CO2).
NB: if the applied volume is too high, many cells will stay and grow out of the matrix.
To determine the perfect number of cells for each hydroscaffold, a range of cell densities should be tested. Don’t hesitate to contact us to get information about cells we have already tested in BIOMIMESYS® in the past.
