NFkB (Nuclear Factor kappa-light-chain-enhancer of activated B cells) is found in almost all animal cell types and is a key player in the inflammatory response. It belongs to the catefory of “rapid-acting” primary transcription factors since it is present in cells in an inactive state, and is quickly activated and is a first responder to harmful cellular stimuli, including pro-inflammatory stimuli.
Figure 1 : Schematic representation of NFkB translocation from cytosol to nucleus.
We can performed NFkB translocation assessment on primary keratinocytes and fibroblastes, as on HacaT cell line.
Figure 2 : Human primary keratinocytes exposed 4 hours to LPS. Nucleus are stained in blue, NFKB in green.
You can multiplex this assay by monitoring pro and anti-inflammatory genes expression and interleukine excretion (RNA-FISH, ELISA, …).
